Efficient Protein Separation with HEPES-Tris Buffer and Advanced Gel Technology

In protein electrophoresis, the HEPES-Tris buffer system plays a vital role in enhancing separation efficiency while reducing the overall electrophoresis time. This system is specifically designed to provide consistent pH levels, making it suitable for both native and denatured protein electrophoresis.

Advanced Gel Composition and Uniformity

The development of acrylamide gels with both fixed and gradient concentrations ensures flexibility in protein analysis. Fixed concentrations, such as 8%, 10%, 12%, and 15%, cater to specific experimental needs, while gradient concentrations (e.g., 4-12% and 4-20%) offer broader separation capabilities. These gels are manufactured using sophisticated perfusion technology, which guarantees uniform polymerization and consistent distribution, crucial for reliable results across different batches.

Plastic Gel Plates and Stability

The adoption of plastic gel plates provides enhanced durability and ease of use during electrophoresis. The application of specialized perfusion technology in these plates ensures that the gels maintain stable performance and consistency, batch after batch. This stability is essential for producing reproducible electrophoresis outcomes.

Applications in Protein Electrophoresis

These gels are versatile and can be used for both native and denatured protein electrophoresis. In native conditions, they effectively separate proteins with an isoelectric point below 7, provided the electrophoresis solution is compatible with the gel system. This compatibility is key to achieving optimal separation results.

Sample Loading and Configuration

The gels are designed with varying well configurations to accommodate different experimental setups. Gels with 10 wells offer a maximum sample capacity of 70 μL per well, while those with 15 wells can handle up to 40 μL per well. This versatility allows for customized experimental designs based on the specific requirements of the research.

Compatibility with Electrophoresis Systems

These gels are compatible with a wide range of mini vertical electrophoresis tanks, including popular systems like the Bio-Rad Mini-Protean series and the Life Technology Novex Mini-Cell. This broad compatibility ensures that laboratories can integrate these gels seamlessly into their existing workflows without the need for significant equipment changes.

Critical Operational Considerations

When performing electrophoresis, especially in native protein analysis, it's essential to ensure that the electrophoresis solution is fully compatible with the gel system. Additionally, structural features like the stacking gel height, typically 1.5 cm, contribute to the overall efficiency and effectiveness of the separation process.

The Precast Protein Plus Gel series utilizes a HEPES-Tris buffer system, offering the benefits of excellent separation performance and significantly reduced electrophoresis time. The Precast Protein Gel plate is made from plastic, and the use of specialized perfusion technology ensures stability and consistency in strip distribution across different batches of Precast Protein Gel, resulting in a stable electrophoresis effect.

The stacking gel height is 1.5 cm, with gradient concentrations ranging from 4-12% and 4-20%, and fixed concentrations available at 8%, 10%, 12%, and 15%. Each concentration gel includes 10 or 15 sample loading wells. The maximum sample loading capacity is 70 μL for 10 wells and 40 μL for 15 wells.

These gels do not contain SDS and are suitable for both natural and denatured protein electrophoresis. When used for natural protein electrophoresis, only proteins with an isoelectric point below 7 can be separated. The electrophoresis solution must be compatible with the gel system. The Yeasen MiniPro® PET 2 mini vertical electrophoresis tank (Cat#80210) is compatible with most mini SDS-PAGE electrophoresis tanks, including the Bio-Rad Mini-Protean (II/3/Tetra System) and Life Technology Novex Mini-Cell (with a special baffle).
